摘  要:采用丫啶橙(AO)荧光染色、流式细胞术和电镜观察非甾体类抗炎药消炎痛诱导人胃癌细胞株MKN28的凋亡。结果发现:经消炎痛作用2h后,MKN28细胞的AO染色阳性率上升,并和消炎痛呈剂量依赖关系,提示消炎病可诱导MKN28细胞凋亡,且凋亡的诱导在2h内即可启动。流式细胞未显示细胞周期亦发生变化,C0/G1期细胞从60.1%下降至28.6%,S期从28.5%增加至35.6%,G2/M期细胞从11.4%增加至35.8%。超微结构的变化为:细胞变圆,细胞核浓缩,染色质越边缘化分布。提示非衡作类抗炎药消炎病可诱导人胃癌细胞株MKN28凋亡,且凋亡的诱导在较短时间内即可完成,并对其机理进行探讨。The effect of the nonsteroidal anti-inflammatory drug (NSAID) indomethacin on apoptosis in human gastic cancer cell line MKN28 was studied with fluorescent acridine orange (AO) staining, flow cytometry (FCM) and electron microscope. After expoaing the MKN28 cells to indomethacin, the resulTS of the AO staining show an increase in apoptosis positive results, depending on the concentration of the indomethacin used. Inducement of apoptosis was triggered within two hours. The number of cdls in the G0/G1 phase of the cell cycle decreased from 60.1 % to 28. 6 %. Cells in the S phase increased from 28. 5 % to 35. 6%, and cells in the G2/M phase increased from 11. 4 % to 35. 8 %, which was demonstrated through FCM. Cells rounded, nuclear pyknosis and chromatin gathered along the nuclear membrane were observed through electron microscope. These findings suggest that indomethacin causes apoptosis in the MKN28 cell line and triggers apoptosis in a short time. The mechanism by which indomethacin works has also been explored.
