摘 要:目的应用噬菌体表面展示技术寻找原因不明性肝炎血清蛋白成分在肝细胞表面的结合蛋白,探讨原因不明性肝炎发病机制。方法将原因不明性肝炎患者血清蛋白作为固相筛选分子,对噬菌体正常人肝细胞T7 cDNA文库进行4轮生物筛选,经噬斑PCR测序,获得原因不明性肝炎血清蛋白结合蛋白的cDNA序列,将其在GenBank中搜索,比对可能与原因不明性肝炎血清蛋白相结合的蛋白。结果筛选出已有功能研究的蛋白9个和未知功能蛋白3个。结论发现了12种可能与原因不明性肝炎血清蛋白结合的肝细胞表面蛋白,为进一步研究原因不明性肝炎的发病机制提供基础。[Objective] To search for liver surface inding protein of serum protein composition in idiopathic hepatitis by phage display technique,and investigate the pathogenesis of idiopathic hepatitis. [Methods] Serum protein of idiopathic hepatitis, as the target protein, was screened by the T7 select phage human liver cDNA li- brary and positive clones were selected 4 times. After polymerase chain reaction of bacteriophage plaque, the DNA sequences of screened clones were analyzed, and the amino acid sequence of target protein was compared in protein sequence database from BLAST in GenBank. [Results] 9 function protein and 3 unknown function pro- tein were obtained. [Conclusion] 12 liver surface proteins of idiopathic hepatitis-binding protein were obtained by T7 cDNA phage display system, which provide basis for farther study the nosogenesis of idiopathic hepatitis.
