摘  要:建立了高效液相色谱-二极管阵列检测法研究内酯式他汀的水解行为,同时测定洛伐他汀和辛伐他汀及有关物质。色谱条件为:Waters Symmetry C18色谱柱(4.6mm×150mm,5μm);乙腈-磷酸二氢钾缓冲液(60∶40)为流动相;流速1.0mL/min;检测波长为238nm。结果表明,洛伐他汀和辛伐他汀在适当条件下可以完全水解成相应的开环酸。确定了洛伐他汀和辛伐他汀及有关物质的线性范围。本法快捷准确,可作为辛伐他汀原料药生产过程及洛伐他汀发酵液的质量控制方法。To investigate the hydrolysis of two lactone form statins, high performance liquid chromatographydiode array detection (HPLC-DAD) method was developed to determine Iovastatin and simvastatin and their related substances simultaneously. Waters Symmetry C18 column(4.6mm×150mm, 5μm) was used and the mobile phase was a mixture of acetonitrile-sodium dihydrogen phosphate (60:40), flow rate 1.0mL/min, detected at 238nm. Under proper conditions, Iovastatin and simvastatin could be hydrolyzed to their corresponding β-hydroxy acids, respectively. The linear ranges of four related substances were determined by HPLC-DAD method.The method is simple, quick and accurate, can be used to the quality control of synthetic simvastatin as well as the lovastatin fermentation process.
