摘  要:目的:利用肌萎缩侧索硬化症的脊髓片培养模型,观察胶质细胞源性神经营养因子(GDNF)对运动神经元的保护作用。方法:取出生后8天乳鼠的腰段脊髓组织切片做脊髓器官型培养,随机分为对照组、模型组、不同浓度GDNF组(1ng/ml、5ng/ml和50ng/ml),倒置显微镜观察脊髓片形态变化,SMI-32免疫组化观察前角运动神经元数目的变化。结果:对照组体外生长良好,而模型组生长不良,SMI-32阳性细胞较对照组减少,GDNF各组脊髓运动神经元存活数目较模型组显著增多,且具有量效关系。结论:GDNF能保护运动神经元免受谷氨酸兴奋毒性的损伤。Objective: Investigate the protective effects of glial cell derived neurotrophie factor (GDNF) on injured motor neurons mediated with glutamate. Method: Organotypie spinal cord cultures were made in 8-day-old rats. The slices were divided into 5 groups: control group, model group and GDNF 1ng/ml、5ng/ml and 50ng/ml group. We inspected the morphologie changes with inverted microscope and calculated the number of spinal motor neurons with SMI-32 immunohistoehemieal staining. Result: The spinal cord slices of GDNF groups had good morphology similar to those of the control group. The number of SMI-32 positive α motor neurons decreased in model group compared with control group while GDNF groups dose-dependently increased compared with model group. Conclusion: GDNF could protect motor neurons from the glutamate exeitotoxieity.
