大骨鸡血清淀粉酶同工酶谱及其活性的测定

摘　　要：以纯种大骨鸡为研究对象，采用垂直板聚丙烯酰胺凝胶电泳法对大骨鸡血清淀粉酶同工酶谱进行分析，并利用比色法对淀粉酶活性进行测定。结果表明：①大骨鸡血清淀粉酶有2种同工酶Amy-1和Amy-3，其中Amy-1有AA、AB和BB3种表型而呈现多态性，表型频率分别为10.42％，80.21％，9．37％，AB为优势表型，Amy-1^A和Amy-1^B基因型频率分别为50.53％。49．47％。（多大骨鸡血清淀粉酶活性平均为（690．48±98．35）U／L，AB型大骨鸡血清淀粉酶活性高于AA和BB型。Purebred Dagu chickens were used to study the spectra-isozyme and activity of serum amylase with vertical polypropylene gel electrophoresis and spectrophotometry,respectively. The results were as follows：there were two isozymes of serum amylase including Amy-1 and Amy-3. Amy-1 existed the polymorphism in Dagu chicken and there were three kinds of types that were AA,AB and BB and AB was the advantageous type. The frequency of phenotype of AA, AB and BB were 10.42%,80,21%,9.37%,and the frequency of gene type of Amy-1A and Amy-1s were 50.53% and 49.47% respectively. The average activity of serum amylase was （690.48±98.35）U/L. And the activity of serum amylase in the Dagu chicken with AB phenotype was significantly higher than that in the chicken with AA and BB phenotype.
