摘  要:目的通过应用GABAB受体阻断剂Saclofen探讨金属铝对海马CA3区诱发电位的抑制作用是否与GAB-AB受体有关。方法应用单个电脉冲(波宽0.15ms,强度30V)刺激穿通纤维(PP),在海马CA3区记录诱发的群体峰电位(PS)。在CA3区微量注射AlCl3和Saclofen,观察它们对PS幅值的影响。结果①0.5mol/LAlCl3注入CA3区后,PS幅值较注药前显著减小(P<0.01或P<0.05)。②25μmol/L的Saclofen易化CA3区PS的幅值(P<0.01或P<0.05)。③50μmol/L Saclofen显著抑制CA3区PS的幅值(P<0.01或P<0.05)。④25μmol/L的Saclofen(P<0.05)和50μmol/L的Saclofen(P<0.01或P<0.05)均加强AlCl3的抑制效应。结论GABAB受体可能部分介导了铝对海马CA3区PS的抑制作用。Objective To investigate whether the inhibitory effect of aluminum on the evoked potential in hippocampal CA3 area Was related to GABAB receptor with GABAB antagonist Saclofen. Methods By using extracellular recording technique, the population spike (PS) was recorded in hippocampal CA3 area by stimulating perforant path (PP) with a single pulse (duration 0.15 ms, intensity 30 V). The effects of Saclofen and(or) aluminum chloride (A1C13) microinjected into the hippocampal CA3 on the PS were observed. Results O After microinjection of AlCl3 (0.5 mol/L, 1μl) into CA3, the amplitude of PS in CA3 was significantly decreased (P〈0.05 or P〈0.01) as compared with that before microinjection of AlCl3. ② 25 μmol/L Saclofen could enhance the amplitude of PS (P〈0.05 or P〈0.01).③ 50μmol/L Saclofen could significantly depress the amplitude of PS in CA3 area (P〈0.05 or P〈0.01).④ Both 50μmol/L (P〈0.05 or P〈0.01) and 25μmol/L (P〈0.05) Saclofen reinforced the inhibitory effect of AlCl3 on PS recorded in CA3. Conclusion The inhibitory effect of AlCl3 on PS in CA3 area might be partly mediated by the GABAB receptor.
