摘  要:目的:通过对正常SD大鼠及肝功能衰竭模型、肝纤维化模型肝组织中骨形态发生蛋白7(bone morpho-genetic protein-7,BMP-7)表达的分析,初步探讨BMP-7在肝脏炎症和肝细胞再生过程中可能发挥的作用。方法:分别采用腹腔注射四氯化碳或D-氨基半乳糖的方法构建SD大鼠肝纤维化模型及肝功能衰竭模型。采用RT-PCR方法研究BMP-7 mRNA的表达,并对4例正常SD大鼠、11例肝功能衰竭模型、3例肝纤维化模型中BMP-7 mRNA的表达进行实时荧光定量RT-PCR分析。结果:BMP-7在正常SD大鼠肝脏有表达,BMP-7/β-actin在正常大鼠肝组织(0.449±0.154)高于肝功能衰竭模型肝组织(0.044±0.024),低于肝纤维化模型肝组织(3.174±2.525),与后者的差异无显著性。结论:BMP-7在正常SD大鼠肝脏有表达,在肝纤维化模型表达增加,在肝功能衰竭模型表达减少。Objective:To investigate the possible effects of bone morphogenetic protein 7 (bone morphogenetie proteins,BMP-7) on liver inflammation and hepatoeyte regeneration by analysis of its expression in the liver of normal SD rat, model of hepatic fibrosis and model of hepatic failure. Methods:SD rat models with hepatic fibrosis were made through intraperitoneal injection of 40% carbon tetraehloride 0.2 ml per 100 g rat body weight three times perweek for six weeks while models with hepatic failure was obtained 24 hours after intraperitoneal injection of D-galaetosamine 800 1 200 mg per 1 000 g rat body weight. To study the expression in the liver tissues of normal and model SD rat by RT- PCR and gel eleetrophoresis. BMP 7 mRNA expression in the liver of 4 normal SD rat, 3 SD rat models with hepatic fibrosis and 11 SD rat models with hepatic failure was quantitated by real-time RT-PCR. Results: BMP-7 mRNA expressed in the liver of SD rat, BMP-7/β-actin in the liver tissue of normal SD rat (0. 449±0. 154) was higher than that in hepatic failure models(0. 044±0. 024) while lower than that in the hepatic fibrosis models(3. 174±2. 525), but the difference with the latter is of no statistical significance. Conclusion:BMP7 expression in the liver of normal SD rat, increases in hepatic fibrosis model, but decreases in hepatic failure model.
