摘 要:目的观察鞘脂酶激活蛋白(sphingolipid activatorproteins,Saposins)在年龄相关性白内障晶状体上皮细胞的表达。方法收集20例年龄相关性白内障和10例正常透明晶状体的前囊膜(中央5.5~6.0mm直径区域),采用免疫组织化学染色法检测晶状体上皮细胞中鞘脂酶激活蛋白的表达并计算灰度值。结果两组晶状体上皮细胞中均有鞘脂酶激活蛋白的表达。与正常透明晶状体组相比,Saposin A(145.03±11.96)、Saposin C(139.78±7.99)和Saposin D(144.30±8.22)在年龄相关性白内障晶状体上皮细胞的表达量增加(P均<0.01),SaposinB(132.59±16.91)的表达量无明显增加(P>0.05)。结论鞘脂酶激活蛋白可能在晶状体老化和年龄相关性白内障形成过程中起重要作用。Objective To investigate the expression of sphingolipid activator proteins (Saposins) in the lens epithelium of age-related cataract. Methods The central 5.5mm to 6.0mm diameter of the anterior lens capsule was collected from both age-related cataract (20 cases) and normal clear lenses (10 cases). Immunohistochemistry (streptavidin-peroxidase, SP) was used to detect the gray scale intensity level of the Saposins in the cataractous and normal lens epithelium, and the value of gray in the positive expression was counted and analyzed. Results Saposin A, B, C and D were all detected at significant levels in the lens epithelium of both the age-related cataract and normal clear lens groups. The expression of Saposin A (145.03±11.96), Saposin C (139.78±7.99), and Saposin D (144.30±8.22) in the lens epithelium of age-related cataract was higher than that in the normal clear lens (124.30±11.10, 127.30±10.00, and 134.63±6.47, respectively, all P〈 0.01). The expression of Saposin B (132.59±16.91) in the lens epithelium of age-related cataract did not increase markedly (P 〉0.05) in comparison to the normal clear lens group (131.21 ±15.90). Conclusion The changes in the expression of Saposins when comparing normal and eataractous lens epithelium indicate that the Saposins family may play a role in lens aging and cataract formation.
