清金得生片对人肺腺癌细胞PCNA及p53蛋白表达水平的调节

摘　　要：目的探讨清金得生片抗肺癌的作用机制。方法运用药物血清和细胞药理学方法,用ELISA法测定清金得生片对培养上清液中p53蛋白含量的影响;免疫组化(SABC)法检测清金得生片对人肺腺癌细胞PCNA、p53蛋白表达水平的影响。结果清金得生片药物血清作用72 h,培养上清液中p53蛋白含量显著地降低,人肺腺癌细胞PCNA及p53蛋白阳性染色率下调,并呈明显的量效关系。结论清金得生片可抑制肺腺癌细胞的增殖活性和p53基因的突变,从而达到抗肺癌作用。Objective To explore the mechanism of anti-tumor effects of qing jin de sheng tablets on lung cancer. Methods To apply the serum contained medicine and cytopharmacology approach to observe the effects of qing jin de sheng tablets on the levels of p53 protein, which were determined by ELISA method,in upper cultured liquid;on the levels of PCNA and p53 protein expressions,which were determined by SABC method, in human lung adenocarcinoma cells. Results 72 hours after the addition of the serum contained qing jin de sheng tablets,the levels of p53 protein in upper cultured liquid reduced significantly; and the positive dyeing rate of PCNA and p53 proteins reduced in human lung adenocarcinoma cells. The above reductions showed obvious dose-effect relations. Conclusion Qing jin de sheng tablets have anti-tumor effects against lung cancer by inhibiting the proliferation of lung adenoearcinoma cells and the mutation of p53 gene.
