摘  要:目的 从人体肺癌组织分离纯化热休克蛋白70-肿瘤肽(HSP70-肿瘤肽),比较两种层析系统分离纯化的结果。方法 采用快速蛋白液相层析(FPLC)自动系统及简单离子交换层析系统,运用亲和层析、离子交换层析、SDS—PAGE、Western-blot方法从肺癌病人手术切除肿瘤组织分离纯化HSP70-肿瘤肽。结果收集两种方法所纯化蛋白洗脱液,经电泳显示于70KD处有一纯蛋白带,定性鉴定该蛋白确系HSP70肽蛋白。结论两种层析系统均能够纯化出纯度较高的热休克蛋白70-肿瘤肽,两种方法均切实可行,实验室可根据自身条件选择应用。Objective To purify HSP70-tumor peptide from lung tumor tissues of patients and compare the purification of protein with different chromatography systems. Methods With fast protein liquid chromatography (FPLC) and simple ion exchange chromatography system , HSP70-tumor peptide were purified from tumor tissues by the method of affinity chromatography, ion exchange chromatography, SDS-PAGE, and Western-blot. Results After electrophoresis, 70kD molecules were identified and the protein lane in the PVDF membrane were confirmed to be HSP70 with Western-blot. Conclusions HSP70-tumor peptide could be purified from human tumor tissues with different chromatography systems, and either of them could be chosen in laboratory to obtain HSP70-tumor peptide.
