桃基因组DNA的提取及SSR反应体系的建立

摘　　要：针对桃树树种-大久保桃，进行了其基因组DNA的提取，探讨了SSR反应体系中各组分浓度对扩增反应产物的影响．结果表明，采用的提取方法所得的基因组DNA纯度好、片段完整，无明显降解；在25μL体系中的适宜浓度分别为：1×PCRBuffer，TaqDNA酶2U，Mgcl2 2mmo1／L，模板DNA80ng，dNTPs240～360μmol／L，每个引物0．4μmol／L．This paper sudied the breed of peach- okubo and the extract of genomic DNA , effect of elements concentration of SSR reaction system on augment reaction prodution. The result showed that the purity of the extract of genomic DNA was pretty good and the fragment was whole, didn＇ t have obviously degradat; the suitable concentration of 25 μsystem was ： 1× PCR Buffer, Taq DNA polymerase 2 U, Mgcl2 2 mmol/L, template80 ng, dNTPs 240- 360μmol/L, each primer 0.4 μmol/L.
