8-氯-环腺苷酸对人膀胱癌细胞中野生型p53、突变型p53 mRNA表达的影响

摘　　要：目的探讨cAMP类似物8-氯-环腺苷酸（8-Cl-cAMP）对人原代培养的膀胱癌细胞中野生型p53（wtp53）、突变型p53（mtp53）mRNA表达的影响。方法原代培养膀胱癌细胞,运用免疫组化及原位杂交方法检测膀胱癌细胞中wtp53、mtp53 mRNA的表达。结果8-Cl-cAMP处理后的膀胱癌细胞中mtp53 mRNA表达水平明显低于未处理组,而经8-Cl-cAMP处理后的膀胱癌细胞中wtp53 mRNA表达水平则明显高于未处理组,两组相比差异均有统计学意义（P〈0.05）。结论8-Cl-cAMP可抑制膀胱癌细胞中mtp53 mRNA,上调wtp53mRNA表达水平,进而抑制肿瘤细胞的分裂和增殖。Objectiv e To study the effects of 8 - Cl - cAMP on the expression of wtp53, mtp53 gene in bladder carcinoma cells. Methods After human bladder carcinoma cells were cultured primarily in the media containing 10-5 mol/L of 8- Cl -cAMP ,the alteration of expression of wtp53, mtp53 gene were observed by using in situ hybridization. Results The expression of mtp53 mRNA in experimental group were weaker than that in control group, and the expression of wtp53 mRNA in experimental group were stornger than that in control group ,there was significant difference between control group and experimental group （ P 〈 0.05 ）. Conclusion 8 - Cl - cAMP could inhibit the expression of mtp53 gene in bladder carcinoma cells,and enchance the expression of wtp53 gene in bladder carcinoma cells. It indicated that 8 - Cl - cAMP was an effective inhibitor for mitotic and proliferation of tumor cells.
