摘 要:目的探索三七总皂苷对大鼠小肠缺血再灌注损伤的保护作用.方法采用肠系膜上动脉夹闭-松夹方式复制大鼠小肠缺血再灌注损伤模型.实验动物分4组,即模型对照组(缺血再灌注组)、三七总皂苷200mg/kg和400mg/kg实验组、假手术组.实验组在建立肠缺血再灌注损伤模型12h前灌胃给予三七总皂苷预处理.分别采用鲨试验偶氮显色法和酶联紫外分光光度法检测血浆LPS和D-乳酸.取肝、脾、肠系膜淋巴结及血液分别作细菌培养.取回肠一段,按Chiu氏分级法进行病理组织学的分类评价.结果模型对照组细菌培养阳性数显著增加,而实验组的明显减少.两种剂量的三七总皂苷均明显降低血浆LPS浓度,400mg/kg的三七总皂苷降低血浆D-乳酸浓度,而200mg/kg剂量则无明显影响.病理形态学检查结果显示,三七总皂苷明显改善缺血再灌注引起小肠的病理性改变.结论三七总皂苷对小肠缺血再灌注损伤有较好保护作用.Objective To study the effect of Panax notoginsenoside on intestinal ischemia -reperfusion (I - R) injury in SD rats. Methods The I - R rat model was prepared by occluding the superior mesenteric artery (SMA) for 1 h, and followed by reperfusion for 1 h. SD rats were divided into four groups: intestinal ischemia - reperfusion group, Panax notoginsenoside - pretreated two groups (200 mg/kg and 400 mg/kg), and Sham - operated group. D -lactate and endotoxin (lipopolysaccharide, LPS) levels were determined, and the I -R intestine changes were examined by histology. Results The levels of LPS and D -lactate in Panax notoginsenoside -pretreated groups were lower as compared with control group. The bacterium positive rates in Panax notoginsenoside - pretreated groups were lower than control group. Intestine histological change in control group was more significant than that in Panax notoginsenoside - pretreated groups. Conclusion Panax notoginsenoside showed marked preventive effects against intestine I -R injury in rats.
