摘 要:目的寻找一种有效获得神经干细胞单细胞的方法。方法运用胰蛋白酶,EDTA和不同浓度酵素对人胚神经干细胞球进行消化,免疫细胞化学染色鉴定及其分化。结果运用一定浓度的酵素可以获得大量成活的单个神经干细胞。而运用胰蛋白酶,EDTA消化后成活细胞少。结论运用一定浓度的diapase消化神经干细胞球是可获得大量成活的神经干细胞单个细胞的方法。[Objective] To investigate an effective way to isolate neural stem cells sphere(neurospheres). [Methods] .Neurospheres were dissociated by digestion with trypsin, EDTA and different dose of dispase, and immunohistochemistry techniques were used for the neurospheres and their differentiation. [Result] Lots of viably single neural stem cells can be obtained by a certain of dispase, then the neurospheres with trypsin and EDTA were damaged and few viably cells obtained. [Conclusion] It's an ideal mothod by using dispase to digest neurosphere for single neural stem cell.
