摘 要:以凯特、新世纪以及凯特×新世纪杏杂交后代群体(F1)株系为试材,进行自花授粉,利用荧光显微镜观察授粉后花粉管生长动态,进行S基因特异PCR扩增,并对花柱中RNA进行RT-PCR。研究结果表明:自交亲和的花粉管能顺利进入子房,而自交不亲和的花粉管顶端膨大呈球形,停止向下生长;发现凯特×新世纪的杂种后代群体中有4种S基因型,杂种后代相同S基因型中存在自交亲和与自交不亲和的分化;田间杂交坐果率符合S基因检测的预测。Katy(SC),Xinshiji(SI) and their FI hybrid progenies in apricot were used in this study. Pollen tubes traveling along the style had been observed in Katy and Xinshiji after self-pollinated by fluorescence that pollen tubes traveling in Katy could grow into ovary and most in Xinshiji could stop growing with tip expanded into ball. DNA was amplified through PCR using AS1 Ⅱ and AnnyC5R as primers designed according to the conserved (CI, C5 ) sequences of Rosaceae S-RNases. Three S-RNase genes, P. a S8 from Katy and P. a S9, P. a S10 from Xinshiji, were detected and cloned. S-genotype were identified S9Se (se means self-compatibility S-gene absent in style) for hybrids No. 35,52, S8S9 for 42,46,and S8S10 for 50,55 .Self-incompatibility among the same S-genotype hybrid seedlings was not contained and their cross-pollinated fruiting rates in field were confo rmed to the results expected by the S-genotypes.
