摘  要:目的探讨银杏提取物(EGb)对体外培养的人类成纤维细胞的抗氧化作用。方法分别于常态氧(氧浓度为21%)及高氧(氧浓度为40%)环境下培养人胚肺成纤维细胞(MRC-5),并给予不同浓度(0,7.5,15,30,60μg/ml)的EGb,以400μmol/L的α-苯基-N-4-丁基硝酮(PBN)为阳性对照,观察不同条件下细胞的生长曲线,并用流式细胞仪测定不同时期、不同条件下MRC-5细胞内活性氧(ROS)的含量。结果常规培养条件下,MRC-5在不同浓度EGb下的生长曲线无明显差别,不同时期测定的ROS含量与阴性或阳性对照比较,总体上无显著差异;而在高氧环境中,使用EGb的MRC-5细胞生长绝对数明显比空白对照多,且呈现EGb浓度越高,细胞绝对数越多、不同时期ROS含量越低、衰老细胞越少的趋势。结论银杏叶提取物对慢性氧化应激下的MRC-5有保护及抗衰老作用。Objective To explore the antioxidative and anti-aging effect of extract of ginkgo biloba (EGb) on human fibroblasts in vitro. Methods Human embryo pulmonary fibroblasts ( MRC-5 ) were cultured under circumstance of normoxia ( oxygen concentration 21% ) and hyperoxia (oxygen concentration 40% ) respectively and treated with different concentrations of EGb (0, 7. 5, 15, 30, 60 μg/ml, respectively). The well with 400 μmol/L phenyhertbutylnitrone (PBN) served as positive control. The growth curves of MRC-5 were recorded and intracellular content of reactive oxygen species (ROS) of MRC-5 were measured in different conditions and time points. Results As a whole, under normoxia, neither growth curves nor ROS contents were significant different in various concentration of EGb. While in the circumstance of hyperoxia, the absolute number of MRC-5 in wells treated with EGb were greatly higher than those in untreated ones, with a trend of the more EGb given, the better MRC-5 grew, the less intracellular ROS content and the less senescent cells in different time points. Conclusions EGb has a protective and anti-aging effect on MRC-5 fibroblasts under chronic mild oxidative stress in vitro.
