摘  要:目的探讨cAMP类似物8-Br-cAMP对人原代培养的膀胱癌细胞中H-ras、野生型P53(wtP53)mRNA表达的影响。方法原代培养膀胱癌细胞,运用原位杂交、斑点印迹方法检测膀胱癌细胞中H-ras、wtP53mRNA的表达。结果8-Br-cAMP处理后的膀胱癌细胞中H-ras mRNA表达水平明显低于未处理组,而经8-Br-cAMP处理后的膀胱癌细胞中wtP53mRNA表达水平则明显高于未处理组,2组相比差异均有统计学意义(P<0.05);两种方法检测处理前后的H-ras、wtP53mRNA表达数值差异与显著性(P>0.05)。结论8-Br-cAMP可抑制膀胱癌细胞H-ras mRNA,上调wtP53mRNA表达水平。Objective To study the effects of 8 - Br - cAMP on the expression of H - ras, wtP53 gene in bladder carcinoma cells. Methods After human bladder carcinoma cells were cultured primarily in the media containing 10 - 5mol/L of 8 - Br - cAMP, the alteration of expression of H - ras,wtP53 gene were observed by using in situ hybridization and dot blot. Results The expression of H-ras mRNA in experimental group were weaker than that in control group, and the expression of wtP53 mRNA in experimental group were stronger than that in control group, there was significant difference between control group and experimental group ( P 〈 0.05 ). There was not significant difference between the data by using in situ hybridization and the data by dot blot( P 〉 0.05 ). Conclusion 8 - Br - cAMP could inhibit the expression of H - ras gene in bladder carcinoma cells, and enchance the expression of wtP53 gene in bladder carcinoma cells.
