摘 要:目的 研究HIF-1α、BFGF、BFGFR在大肠腺癌中的表达和相互作用,以及它们与大肠腺癌生物学行为的关系。方法 采用免疫组化SP法检测了60例手术切除的大肠腺癌及癌旁组织,20例正常大肠粘膜组织中的HIF-1α、BFGF、BFGFR表达。结果 HIF-1α、BFGF、BFGFR在大肠腺癌中的表达阳性率分别为61.7%、65.0%、58.3%。而癌旁组织的表达率分别为10.0%、13.3%、11.7%。HIF-1、BFGF、BFGFR在大肠腺癌组织与癌旁组织中表达有显著性差异(P〈0.05)。正常肠黏膜中未检出HIF-1α、BFGF、BFG-FR。HIF-1α、BFGF、BFGFR在大肠腺癌中表达与性别、年龄、肿瘤大小、肿瘤位置和分化程度无关(P〉0.05)。但与肿瘤Dukes分期具有显著性差异(P〈0.05)。大肠腺癌组织中HIF-1α、BFGF与BFGFR具有两两相关性(P〈0.05)。结论 大肠腺癌组织中的HIF-1α、BFGF、BFGFR表达水平增高,参与了大肠腺癌的发生发展过程,与大肠腺癌的预后有关;在大肠腺癌发生发展中HIF-1α、BFGF与BFGFR可能起到协同作用,共同促进肿瘤的发生。Objective To study the expression and interaction of Hypoxia inducible factor - 1 ( HIF - 1 ), basic fibroblast growth factor (BFGF) and basic fibroblast growth factor receptor (BFGFR) in colorectal cancer and the relationship between both HIF - 1 ,BFGF and BFGFR expression and biology behavior of colorectal cancer. Methods 60 intestinal biopsy specimens were taken from patients with colorectal cancer and 20 normal intestinal mucosal were taken from irritable bowel syndrome , expression of HIF-1α and BFGF,BFGFR were detected by immunohistochemical staining( SP method). Results Positive rates of HIF-1α,BFGF and BFGFR in colorectal cancer tissues were 61.7% ,65.0% and 58.3% respectively, Positive rates of HIF-1α,BFGF and BFGFR in para -cancer tissues were 10.0% ,13.3% and 11.7% respectively, HIF-1α, BFGF and BFGFR were not detected in normal intestinal mucosal. Positive rates of HIF-1α,BFGF and BFGFR were significant difference between in colorectal cancer tissues and in para - cancer tissues( P 〈 0.05 ). there were no correlations between gender,age,tumor size,tumor position, cell differentiation and expression of HIF-1α,BFGF and BFGFR in colorectal cancer tissues (P 〉 0.05 ). There were significant correlation among the expression of HIF-1α, BFGF and BFGFR and Dukes stage ( P 〈 0.05 ), furthermore, there were relativity between HIF -1α and BFGF,BFGFR in colorectal cancer tissues. Conclusions Excessive expression of HIF -1α,BFGFand BFGFR in colorectal carcinoma tissues may participate carcinogenesis and development, HIF -1α,BFGF and BFGFR participate in genesis of carcinogenesis of colorectal carcinoma together.
