摘  要:目的:探讨孕期膳食锌缺乏对胎盘、母鼠小肠及肝脏中氨基酸转运蛋白Slc7a9 mRNA表达的影响。方法:27只SD孕鼠随机分为正常对照组(ZC)、锌缺乏组(ZD)及对喂组(PF)。分别于孕9.5d、孕13.5d和孕17.5d取孕鼠血清、肝脏、小肠及胎盘,以半定量聚合酶链反应测定Slc7a9mRNA及肝脏金属硫蛋白1(MT-1)mRNA的相对表达量,以原子吸收法测定血清锌浓度。结果:孕9.5d和孕13.5d时,ZD组血清锌浓度和母鼠肝脏MT-1mRNA的相对表达量显著低于PF组和ZC组。各时间点ZD组胎盘和小肠中Slc7a9mRNA相对表达量均显著低于ZC组和PF组,孕17.5d时PF组与ZC组胎盘之间差异显著。孕9.5d、孕13.5d时PF组与ZC组小肠之间差异显著。各时间点ZD组和PF组孕鼠肝脏Slc7a9mRNA的相对表达量与ZC组相比均显著减低。结论:孕期膳食锌缺乏显著下调胎盘及母鼠肝脏和小肠Slc7a9mRNA的表达。Objective: To investigate the effect of dietary zinc deficiency on the expression of Slc7a9 mRNA in placenta, liver and small intestine of rats during pregnancy. Methods: Twenty-seven healthy pregnant SD rats were randomly divided into three groups: normal control (ZC), zinc deficient (ZD) and pair-fed (PF) groups. Placenta, maternal small intestine and liver were taken at gestational D9.5, D13.5 and D17.5 respectively. The expression of Slc7a9 and MT-1 mRNA was examined by RT-PCR and serum zinc concentrations were measured by atomic absorption method. Results: At gestational D9.5 and D13.5, the zinc concentrations and the relative expression of MT-1 mRNA in the maternal livers in group ZD were significantly lower than those in groups PF and ZC, respectively. At each time point, the expression of placental and intestinal Slc7a9 mRNA in group ZD was significantly lower than those in groups PF and ZC respectively. There was significant difference of placental Sic 7a9 between group PF and ZC at D17.5 and of intestinal Sic 7a9 between group PF and ZC at D 9.5 and D13.5. The relative expression of Slc7a9 mRNA in liver in group ZD was significantly lower than that in groups PF and ZC at each time point. Conclusion: The expression of Slc7a9 mRNA in placenta, liver and small intestine of pregnast rats was significantly downregulated by dietary zinc deficieny during pregnancy.
