摘  要:目的评估二十碳五烯酸(EPA)联合表阿霉素(EPI)对人胃癌细胞株MGC-803的作用。方法采用MTT法检测各种药物单用和联合应用对细胞的作用,电镜观察细胞超微结构的变化,等效线图解法判断两药合用效果,流式细胞仪检测细胞周期分布。结果EPA可抑制MGC-803细胞的生长,且呈剂量及时间依赖性(P<0.01);倒置相差显微镜下可见经EPA培养的胃癌细胞核周存在大量异常颗粒,电镜下在核周及胞浆内可见异常电子致密物;EPA对人胚肺成纤维细胞(HPF)生长起促进作用,且呈剂量依赖性(P<0.01);EPA与EPI联合应用对MGC-803细胞生长的抑制具有协同作用(P<0.05);EPA有一定的G0/G1期阻滞作用,EPI有明显的S期阻滞作用,两者可从不同环节抑制细胞增殖。结论EPA对胃癌细胞株MGC-803有抑制作用,相同剂量对正常细胞无抑制作用。EPA与EPI对胃癌细胞的抑制具有协同作用,与单用EPI相比,联合应用可减少EPI的用药剂量。Objective To evaluate the effect of combination of eicosapentaenoic acid (EPA) and epirubicin (EPI) on the human gastric carcinoma cell MGC-803 in vitro. Methods The effects of EPA and EPI were measured by MTT assay , and the interaction between these two agents was evaluated by the isobologram technique of Berenbaum. Morphous of cell was observed by phase-contrast and electron microscope. Flow cytometry was used for cell cycle analysis. Results EPA significantly inhibited the growth of MGC-803 cells in a dose- and time-dependent way ( P 〈 0. 01 ). Numerous abnormal particles were found around the nucleus of MGC-803 cells under phase-contrast microscope, and also many electron-dense material in cytoplasm were found under electron microscope. EPA significantly stimulated the growth of human embryonal pulmonary fibroblast (HPF) dose-dependently (P 〈 0. 01 ). A strong synergism was found between EPA and EPI in MGC- 803 cells. EPA induced G0/G1-phase arrest but without statistical significance ( P 〉 0. 05 ), and EPI signifi- cantly induced S-phase arrest (P 〈 0. 05) in MGC-803 cells. Conclusions EPA can inhibit cell growth in gastric carcinoma cells but not in normal cells. EPA and EPI have synergetic effect in the inhibition of gastric carcinoma cells. Compared with EPI monotherapy, the combination of EPI and EPA can reduce the dosage of EPI.
