摘  要:目的探讨重组人促红细胞生成素(recombinat human erythropoietin,rhEPO)对大鼠视神经(optic nerve,ON)不完全损伤后视网膜神经元凋亡的抑制作用和对视功能修复的影响。方法雄性Long Evans大鼠随机分为实验组和对照组,每组25只。制作ON不完全损伤模型。实验组和对照组按ON夹伤后不同时间分为伤后1d、3d、5d、7d、14d5个时间点。实验组伤后即时、3d、5d、7d,按5000U·kg-1体重腹腔内注射rhEPO;对照组注射等量生理盐水。伤后1d、3d、5d、7d、14d取材。取材前行闪光视觉诱发电位检测。应用TUNEL法检测视网膜神经元的凋亡。结果伤后7d、14d对照组视网膜凋亡细胞密集分布于视网膜节细胞层及内、外核层。实验组7d、14d视网膜凋亡细胞明显减少,视网膜节细胞层的凋亡细胞与对照组相比有非常显著差异(P<0.01)。2组闪光视觉诱发电位潜伏期各时间点组间比较无显著性差异(P>0.05)。伤后1d、3d、5d、7d实验组P1-N2波振幅和对照组相比无显著性差异(P>0.05);伤后14d时有显著差异(P<0.05)。结论rhEPO对ON不完全损伤后视网膜神经元的凋亡具有抑制作用,可以促进视功能的恢复。Objective To investigate the inhibitive effect of recombinant human erythropoietin(rhEPO) on apoptosis of retinal neurons and observe the effect of rhEPO on recovery of visual function afar incomplete optic nerve(ON) injury in rats. Methods Fifty cases male Long Evans rats were randomly divided into test group ( 25 cases) and control group ( 25 cases). The experimental models of incomplete ON injury were established. Two groups were both divided into 5 groups according to the different time pionts( 1 day,3 days,5 days,7 days, 14 days) after incomplete ON injury, rhEPO or normal saline was injected into abdominal cavity of rats immediately and on the 3rd day,5th day,7th day afar ON injury. The rats were sacrificed on the 1st day,3rd day,5th day,7th day and 14th day after ON injury. Before sacrifice,the examination of flash visual evoked potential were performed. The apoptosis of retinal neurons were examined by TUNEL method. Results Apoptotic cells were intensively distributed in ganglion cell layer,inner plexiform layer and inner nuclear layer in retina of control group on the 7th day and 14th day after ON injury.The number of aptopotic cells decreased notablely in test group on the 7th and 14th day after ON injury, the difference of number of apoptotic cells in ganglion cell layer between two groups was significant( P 〈 0.01 ), In every time point afar injury, the latency of flash visual evoked potential had no difference among these groups(P〉0.05). On the 1st day,3rd day,5th day and 7th day after injury,the difference of amplitude of P1-N2 wave in two groups was not significant ( P 〉 0.05 ), but on the 14th day, which was significant ( P 〈 0.05 ). Conclusion rhEPO can effectively inhibit the apoptosis of retinal neurons and promote the recovery of visual function after incomplete ON injury.
