摘 要:目的:细胞质在细胞内的代谢过程、信号转导等功能中起着重要的作用,以体外培养的人体肝癌细胞和正常肝细胞为研究对象,运用蛋白质组学的研究方法,对比分析肝癌细胞与正常肝细胞细胞质蛋白质的差异表达情况,为肿瘤发生机制的阐释提供信息。方法:体外培养的细胞(包括肝癌细胞、正常肝细胞)以超离心分离细胞质,双向电泳分离细胞质蛋白质,图像分析肝癌细胞与正常肝细胞的差异表达蛋白质,MALDI-TOF-MS鉴定差异蛋白质。结果:在肝癌细胞与正常肝细胞的对比分析中筛选出10个差异表达蛋白质,这些差异蛋白质涉及到细胞的能量代谢、蛋白质合成、调亡调控等方面。结论:细胞癌变后细胞质中蛋白质的表达发生了许多改变,可为肿瘤发生机制的阐释提供有价值的信息。Objective: To compare the difference of cytosol protein expression between hepatoma cells and normal hepatocytes, and so as to provide contribution to the explanation of tumorigenesis and action mechanism of epirubicin. Methods: Cultured ceils ( including hepatoma cells and normal hepatocyte ) were subjected to centrifugation for the fractionation of cytosol, and the cytosol proteins were separated by using two dimensional gel electrophoresis. The expression-ahered protein spots were detected with imaging analysis, and the proteins were identified with MALDI-TOF-MS. Results: 10 expression-altered proteins were identified between the hepatoma ceils and normal hepatocytes, and were shown to be involved in energy generation, protein biosynthesis, and apoptosis regulation of the ceils. Conclusions: There occur many changes of cytosol protein expression in hepatoma cells after tumorigenesis, and the results of this study are helpful for us to understand tumorigenesis to some extent.
