摘 要:目的研究实验性肾动脉狭窄性高血压大鼠主要靶器官的细胞凋亡状况以及四逆汤对细胞凋亡的干预作用。方法健康雌性Wistar大鼠24只,随机分为正常对照组、肾性高血压组和四逆汤治疗组,每组8只。双肾动脉夹闭法建立肾性高血压动物模型。采用TUNEL法检测心脏和肾脏组织的细胞凋亡水平。结果肾性高血压组大鼠心、肾阳性细胞计数均显著高于正常对照组及四逆汤治疗组(F=228.24、171.75,q=16.26~29.03,P<0.01),四逆汤治疗组阳性细胞计数与正常对照组相比较差异仍有显著意义(q=7.26、9.68,P<0.01)。结论肾性高血压大鼠心、肾细胞凋亡增多,四逆汤对细胞凋亡有一定的抑制作用。Objective To investigate apoptosis of the target organs of experimental hypertensive rats caused by renal artery stenosis and the interventional effects of Sinitang on the apoptosis. Methods Twenty-four female healthy Wistar rats were evenly divided into normal control group, renal hypertensive group, and Sinitang-treated group. The hypertensive model was established by clipping the renal arteries. Sinitang was given to the hypertensive rats and Sinitang-treated rats. The apoptosis of renal and heart tissues of the rat were detected by terminal deoxynucleotidyl transferase-mediated dutp-biotin nick end labeling (TUNEL) . Results The positive cell counting in renal and heart tissues of the rat in hypertensive group was significantly higher than that in the control and Sinitang groups (F=228.24,171.75,q=16.26-29. 03;P〈0.01), the difference of positive cell counting between Sinitang-treated and normal control groups was also significant (q=7.26,9.68;P〈0.01). Conclusion Apoptosis in the kidney and heart of renal hypertensive rats increases, and Sinitang might inhibit their apoptosis.
