摘 要:研究了甲基橙法测定ε-聚赖氨酸的试验条件,得出结论:甲基橙从水中重结晶后,溶于pH6.6、0.1 mol/L磷酸钠缓冲液中,终浓度为1 mmol/L;将2 mLε-聚赖氨酸溶液与2 mL 1 mmol/L甲基橙溶液混合;混合物在30℃条件下剧烈振荡30 min,4 000 r/min离心15 min;取1 mL上清液稀释50倍,在465 nm波长下测吸光度,计算ε-聚赖氨酸含量.The conditions of detecting ε-polylysine (ε-PL) concentration by the methyl orange were studied in the paper. The results were as follow. After recrystallization from water, methyl orange was dissolved in 0.1mol/L sodium phosphate buffer (pH6.6) at a final concentration of 1 mmol/L. Then 2ml ε-PL solution was mixed with 2ml methyl orange solution. The mixtures were vigorously reacted on a reciprocal shaker at 30℃ for 30min and then centrifuged for 15min at 4000 r/min. The absorbance of the supernate diluted 50-fold was read at 465nm and the ε-PL content was calculated.
