摘 要:应用透射电镜和扫描电镜分别研究油菜游离小孢子培养后细胞的超微结构和胚状体的形态。单核晚期小孢子经培养后,具胚状体发生能力的细胞中央液泡消失,积累淀粉,含丰富细胞器。细胞分裂成二细胞至多细胞原胚后,细胞器更为丰富,但淀粉减少。在胚状体群体中,存在大量异形胚,包括单子叶、三子叶和多子叶,连体孪生胚、连体三胚和其它畸形小胚。Electron microscopes were used to study ultrastructural feature of the microspore invivo and in Vitro and the embryoid shape of isolated microspore culture in Brassica napus.The central vacuole began to disappear,and cytoplasm with abundant organelle accumulated starch in some of microspores having embryogenic ability on transfer to culture. After the cells divided into two celled and multicellular proembryoids,the organelleincreased while the starch grains decreased. Many abnormal embryoids such as one ormore than two cotyledons,twin and triple embryoids from single microspore were observed.
