摘 要:目的:研究银杏叶提取物对肝星状细胞增殖及细胞因子TGF-β1、CTGF mRNA及蛋白表达的影响,以探讨银杏叶提取物抗肝纤维化的可能机制。方法:用不同浓度(0、1、10、100、500mg/L)的银杏叶提取物处理HSC-T6,用MTT及流式细胞仪检测其对肝星状细胞增殖及细胞周期的影响,用RT-PCR及Western blotting检测培养24h及48h后各组细胞中TGF-β1、CTGF mRNA及蛋白的表达。结果:银杏叶提取物10、100、500mg/L可抑制肝星状细胞的增殖,与空白对照组相比,G0/G1期DNA含量逐渐增加(P<0.05或P<0.01),S期DNA含量则逐渐降低(P<0.01),增殖指数(PI)逐渐降低(P<0.05或P<0.01);24h各组TGF-β1mRNA及蛋白表达较对照组分别低(28±4)%、(39±4)%、(45±3)%(P<0.01);(27±4)%、(37±4)%、(41±3)%(P<0.01);48h组各组TGF-β1mRNA及蛋白表达较对照组分别低(35±4)%、(49±3)%、(54±3)%(P<0.01);(33±4)%、(48±3)%、(52±3)%(P<0.01);4h各组CTGF mRNA及蛋白表达较对照组分别低(29±6)%、(45±4)%、(56±3)%(P<0.01);(36±4)%、(48±4)%、(49±3)%(P<0.01);48h组各组CTGF mRNA及蛋白表达较对照组分别低(48±6)%、(57±4)%、(69±3)%(P<0.01);(44±6)%、(58±4)%、(62±3)%(P<0.01)。结论:银杏叶提取物可能通过抑制肝星状细胞的增殖,抑制细胞因子TGF-β1、CTGF基因的表达发挥其抗肝纤维化的作用。AIM. To investigate the effect of Ginkgo biloba extract ( EGb761 ) on cell proliferation and expression of transforming growth factor β1 (TGF - β1 ) and connective tissue growth factor(CTGF) in rat hepatic stellate cells ( HSC -T6 ). METHODS: HSC -T6 was cultured with EGb761 at concentration of 1, 10, 100 and 500 mg/L, and without EGb761 as blank control. MTT colorimetric assay was used for detecting the proliferation of HSCs and flow cytometry was used for observing the cell cycles of HSCs under different concentrations of EGb761. The expression of TGF -β1/CT- GF mRNA and protein levels were determined by reverse transcription - polymerase chain reaction ( RT - PCR) and Western blotting after 24 hours and 48 hours. RESULTS : EGb761 inhibited the proliferation of HSCs at the concentration of 10 mg/L, 100 mg/L and 500 mg/L. G0/G1 stage was increased (P 〈0. 05 or P 〈0. 01 ), while the cell count at S stage and PI were reduced (P 〈 0.05 or P 〈 0. 01 ) compared with the blank control. It down- regulated the expression of TGF -β1CTGF mRNA and protein in HSCs. The expressions of TGF- β1mRNA and protein were reduced by (28 ±4)% , (39 ±4)%, (45 ±3)% (P〈0.01); (27 ±4)%, (37 ±4)%, (41 ±3)% (P〈0.01) respectively inEGb761 groups at 24h, while they were decreased by (28±4)%, (39±4)%, (45 ±3)% (P〈0.01); (27±4)%, (37±4)%, (41 ± 3 ) % ( P 〈 0. 01 ) respectively in EGb761 groups at 48 h. The expression of CTGF mRNA and protein in HSCs were lowed by (29 ±6)%, (45 ±4)%, (56 ±3)% (P〈0.01); (36 ±4)%, (48±4)%, (49 ±3)% (P〈0.01) respectively in EGb761 groups at 24 h, while they were reduced by (48 ±6)% , (57 ±4)%, (69 ±3)% (P〈0. 01) ; (44 ± 6 ) % , ( 58 ± 4 ) %, ( 62 ± 3 ) % ( P 〈 0.01 ) respectively in EGb761 groups at 48 h.
