摘  要:用Northern杂交、免疫细胞化学和电镜技术,以垂体ACTH瘤细胞系(AtT20)为对照,从转录、翻译及亚细胞水平检测了5种垂体外不同病理组织来源的肿瘤细胞系ACTH基因表达,包括属于APUD系统的两种人小细胞肺癌(SCLC-H69和SCLC),3种非APUD系统肿瘤:肺腺癌(AGZY83-α、低分化胃粘液癌(MGC-803)和肝癌(Bel-7402)。结果证实,(1)ACTH基因表达不仅存在于APUD系统肿瘤,而且也存在于非APUD系统肿瘤,如AGZY83-α和MGC-803。(2)不同细胞系中POMC(ACTH前体)转录的mRNA表达长度不同(800bp、1200bp、1400bp)。支持激素类物质合成在组织中普遍存在的假说。The pathogenesis of ectopic ACTH syndrome is still puzzling. In this study, Northern blot hybridization, immunocytochemistry and electron microscopy were used to examine ACTH gene expression in five extra pituitary tumor cell lines, including two human small cell lung cancer cell lines (SCLC H69 and SCLC) which belonged to APUD system and three non APUD human carcinoma cell lines (AGZY83 α, MGC 803 and Bel 7402), and the mouse pituitary ACTH secreting tumor cell line (AtT20) as a control. The results showed that, (1) ACTH was produced not only in APUD tissues but also in some non APUD tissues, such as lung adenocarcinoma(AGZY83 α), low differentiated mucoid adenocarcinoma of stomach (MGC 803). (2) Pro opiomelanocotin (POMC) mRNA expressing in different cell lines had different lengths(800bp, 1 200bp, 1 400bp). This results support the hypothesis of hormone producing in universal tissues, but there is different quality of ACTH gene expression
