摘  要:背景与目的:生物信息学是一门由数学、统计学、计算机科学与生物医学交叉结合的新兴科学,在研究肿瘤差异表达基因方面起到越来越重要的作用。本研究主要应用此方法对乳腺增生症病变组织与乳腺癌组织的基因表达差异进行研究,发现其中一乳腺癌高表达基因c11orf59,对其进行初步的生物信息学分析。方法:使用肿瘤基因组解剖计划(cancer genome anatomy project,CGAP)数据库的数字基因表达演示(digital gene expression displayer,DGED)工具,筛选在乳腺增生症病变组织和乳腺癌组织来源基因表达系列分析(serial analysis of gene ex-pression,SAGE)文库中差异表达的基因,并用生物信息学方法对其中一条功能未知基因c11orf59进行初步分析,预测其基因结构、染色体定位、编码蛋白质的理化性质、亚细胞定位、表达谱、蛋白质结构功能域及在其他物种中的相似蛋白等。最后通过半定量RT-PCR对c11orf59在乳腺增生症病变组织及乳腺癌中的表达进行初步论证。结果:通过上述方法共找出差异表达基因185条,对c11 orf59序列进行了初步的生物学分析,明确了该基因编码蛋白为一核蛋白,可能参与了细胞周期的调控,使其增殖活跃并产生恶变。并初步用实验方法证实了其在乳腺癌中的表达情况。结论:以生物信息学挖掘公共SAGE文库是研究肿瘤基因差异表达的新方法,c11 orf59是一个有意义的乳腺癌分子机理研究靶点。Background and purpose: Bioinformatics plays a more and more important role in the research of the differently expressed genes in cancer. This study was to explore the differently expressed genes between normal breast tissue and breast neoplasms by bioinformatics analysis. Methods: We screened the differentially expressed genes between the serial analysis of gene expression(SAGE) libraries from normal breast tissues and breast neoplasms by using the digital gene expression displayer (DEGD) tool in cancer genome anatomy project ( CGAP), then performed an initial bioinformatics analysis on an unknown gene named c11 or f59 that had not defined the functionality, its gene structure, genome localization, the physical and chemical characteristics of its coding protein, subcellular localization, functional domain and homologue proteins of several species were also clarified, c11 or f59 expression in normal breast tissue and breast neoplasms has been evaluated by semi-quantitative RT-PCR. Results: By using the above methods, 185 differently expressed genes were uncovered. Through efficient bioinformatics analysis, C11 or f59 was fundamentally identified to be a nuclear protein, which may involve in cell cycle regulation and induction of eanceration, c11 or f59 expression could be detected by semi-quantitative RT-PCR. Conclusions: Bioinformatics analysis on multiple SAGE libraries is an efficient and new method to investigate and predict the differently expressed genes in cancer. C11 or f59 has been found to be a potential target for cancer researches through bioinformatics analysis.
