摘  要:【目的】探讨含碱性成纤维细胞生长因子(bFGF)的壳聚糖-Ⅰ型胶原复合支架对人牙周膜细胞(HPDLC)生长和增殖的影响。【方法】MTT法观察含bFGF因子壳聚糖-Ⅰ型胶原复合支架浸出液、不含bFGF因子壳聚糖-Ⅰ型胶原复合支架浸出液以及正常细胞培养液对HPDLC生长和增殖的影响,利用流式细胞仪(FCM)检测上述3种液体对HPDLC细胞周期的影响,并将HPDLC接种于含bFGF因子壳聚糖-Ⅰ型胶原复合支架和不含bFGF因子壳聚糖-Ⅰ型胶原复合支架内,通过免疫细胞组织化学和激光共聚焦显微镜来检测HPDLC在复合支架内的存活和增殖情况。【结果】含bFGF因子壳聚糖-Ⅰ型胶原复合支架浸出液对HPDLC生长和增殖的促进作用明显强于不含bFGF因子壳聚糖-Ⅰ型胶原复合支架浸出液和正常细胞培养液,而后两组间无统计学意义;含bFGF因子壳聚糖-Ⅰ型胶原复合支架浸出液促进HPDLC从G1期进入S期;HPDLC在含bFGF因子壳聚糖-Ⅰ型胶原复合支架内的生长和增殖也明显优于不含bFGF因子壳聚糖-Ⅰ型胶原复合支架。【结论】含bFGF因子壳聚糖-Ⅰ型胶原复合支架可以促进体外培养的HPDLC生长和增殖。[Objective] To investigate the effects of chitosan-type Ⅰ collagen composite scaffold combining with bFGF on HPDLCs growth and proliferation. [ Methods] The effects of leachate of chitosan-type Ⅰ collagen composite scaffold carrying bFGF, leachate of chitosan-type I collagen composite scaffold without bFGF, and normal cell culture fluid on HPDLCs growth and proliferation were studied by MTT. The effects of three kinds of fluids on HPDLCs cell cycle were studied by flow cytometer (FCM). HPDLCs were inoculated into chitosan-type Ⅰ collagen composite scaffold combining with bFGF and chitosan-type Ⅰ collagen composite scaffold without bFGF, then the information about HPDLCs growth and proliferation were studied by immunocytochemistry and laser confocal microscopy. [ Results ] The promotion of chitosan-type Ⅰ collagen composite scaffold combining with bFGF leachate on HPDLCs growth and proliferation was better than those of chitosan-type Ⅰ collagen composite scaffold without bFGF leachate and normal cell culture fluid, there had no difference between the last two groups.The leachate of chitosan-type Ⅰ collagen composite scaffold combining with bFGF promoted more HPDLCs from G1 phase into S phase, and the growth and proliferation of HPDLCs inoculated in chitosan-type Ⅰ collagen composite scaffold combining with bFGF were better than those in chitosan-type Ⅰ collagen composite scaffold without bFGF. [Conclusion] The chitosan-type Ⅰ collagen composite scaffold carrying bFGF can promote the growth and proliferation of HPDLCs in vitro.
