摘  要:在建立以毛白杨85号(Populus tomentosa 85)叶片为外植体的再生系统基础上,从预培养时间、浸染菌液浓度、浸染时间、共培养时间及添加AS(乙酰丁香酮)5个方面,以卡那霉素抗性植株作为转化率的衡量指标,研究了各因素对毛白杨85号遗传转化率的影响,建立了高效的遗传转化系统。筛选的高效转化系统为:农杆菌活化菌液用MS液体培养基稀释10倍,浸染5min,共培养4d。毛白杨85号叶盘转化率可达40.0%。该研究为利用叶盘法开展毛白杨85号基因转化培育抗逆新品种奠定了良好基础。Based on the established leaf explant regeneration system of Populus tomentosa-85, Several factors affecting transformation of P. tomentosa 85 were studied by infecting with Agrobacterium tumefaciens LBA4404, such as pre-cuhure time, the concentration of agroinfection, infection time, coculture time and addition of acetosyringone (AS). Which the rate of kanamycin resistance plantlet was the measure index. An effective protocol in optimized conditions which were used MS dilute activated agroinfection for 10 times, 5minutes for infection and 4 d for co-culture medium for transformation of leaf disc of P. tomentosa 85 was developed and the frequency of transformation reached to 40.0%. The results laid a sound foundation for breeding new resistant species of P. tomentosa 85 gene transformation by means of leaf disc method.
