摘  要:目的:研究氯离子通道对小鼠成牙本质细胞MDPC-23生物学特性的影响。方法:应用MTT、生化分析、荧光染色等方法,观察NPPB阻断氯离子通道之后MDPC-23细胞增殖,ALP活性以及形成细胞外矿化基质的能力等方面的变化。结果:氯离子通道被阻断后,细胞的增殖速度减慢,培养上清中的碱性磷酸酶活性明显增高,而且细胞外矿化结节的形成时间比对照组缩短。结论:阻断氯离子通道可抑制成牙本质细胞MDPC-23的增殖,但对其矿化有一定的促进作用。AIM : To investigate the effects of chloride channels on the biological characteristic of mouse odontoblast-like cells, MDPC -23, in vitro. METHODS: NPPB, a kind of chloride channel blocker, was used to block the chloride channels. The cells proliferation was detected by MTT assay. Biochemical analysis and immunofluo- rescence methods were used to study the effects on the alkaline phosphatase activity (ALP) and mineralization. RESULTS: The cells in treatment group proliferated slower than those in the control group. The ALP activity of the treatment group was higher than that of the control group . There was significant difference of ALP activity between the two groups ( P 〈0.01 ). The cells of treatment group formed mineralized nodus after continuouse cultured for 21 days. However, the cells of control group took 28 days to form the nodus. CONCLUSIONS : These results suggested that the chloride channels may be involved in the proliferation and differentiation of odontoblast - like cells, MDPC - 23.
