摘 要:目的成功建立隔离肺灌注化疗的动物模型。方法建立兔隔离单肺灌注系统,使用阿霉素进行灌注化疗,并应用荧光分光光度法检测双肺及心、肝、肾组织中阿霉素含量及肺湿/干重量比。结果隔离灌注肺的阿霉素含量较对照肺及心、肝、肾组织显著升高;而肺湿/干重量比则无明显差别。结论我们使用的隔离肺灌注系统可在肺组织局部得到较高的化疗药物浓度,且未出现急性肺损伤,使用该模型治疗肺癌及肺转移癌应可取得较好效果。Objective To establish the animal model of isolated lung perfusion.Methods Successfully establish a rabbit model of single lung perfusion system. The lung is isolated and the perfusion is limited between the pulmonary artery and pulmonary vein. Use this model and perfuse with Adriamycin(ADM) .Then examine the ADM level in the tissue of two lungs,heart,liver and kidney by the fluorescence spectrophotometry. Exmine the wet/dry lung weight. Results The ADM level in the isolated lung is extremly higher than that in the contrast lung, heart, liver, kidney. The wet to dry weight of two lungs have no difference. Conclusion Higher drug level can get in the isolated lung without acute lung injury. This animal model is safe and effective.
