摘  要:目的探讨血小板源性生长因子(platelet-derived growth factor,PDGF)和增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)在兔外伤性增生性玻璃体视网膜病变(proliferative vitreoretinopathy,PVR)中表达的动态变化及其相关性。方法采用原位杂交法和免疫组化法分别检测兔外伤性PVR模型PDGF和PCNA表达,染色结果行图像分析。结果伤后PDGF和PCNA表达呈现先升高后降低的动态变化,14d组阳性表达最高,与7d、21d、28d组比较差异有显著性;PDGF和PCNA表达之间有显著相关性(P≤0.01)。结论PDGF参与外伤性PVR增殖细胞的生长调控,并与细胞增殖的动态变化呈高度相关性。Objective To explore the dynamic expression and correlation between PDGF (platelet-derived growth factor) and PCNA (proliferating cell nuclear antigen) in rabbit traumatic proliferative vitreoretinopathy (PVR). Methods Expression of PDGF mRNA in the retina was detected by in-situ hybridization examination, and expression of PCNA in the retina was detected by immunohistochemical analysis. Results of the staining were analyzed with an image analysis system. Results After establishing the animal model, the expression of PDGF mRNA and PCNA protein were first up- regulated and then down-regulated. The peak value was found in the 14d group, which significantly differed from the values in the 7d group, 21d group and 28d group. There was a significant correlation between PDGF and PCNA expression. Conclusion PDGF contributes to the regulation of proliferative cells in traumatic proliferative vitreoretinopathy (PVR), and is highly correlated with dynamic changes in cell proliferation.
