缺氧预适应对小鼠海马脑片LTP的影响

摘  要:目的:观测缺氧预适应对小鼠海马脑片LTP的影响。方法:小鼠缺氧0次(H0)、1次(H1)、4次(H4)后取海马组织,制备成离体海马脑片,刺激海马CA3区Schaffer侧枝,在CA1锥体细胞层记录各组LTP。结果:H1组海马CA1区LTP诱出率最低,H0、H4组诱出率均高于H1组。H0和H4组小鼠海马脑片在高频强直串刺激后PS增幅的百分率和fEPSP斜率增幅的百分比变化均显著高于H1组(P<0.01)。H0组与H4组差异无统计学意义(P>0.05)。结论:缺氧抑制LTP的诱导,缺氧预适应可以恢复对LTP的诱导,突触可塑性的变化可能参与缺氧预适应小鼠脑保护。Objective :To study the effect of hypoxia preconditioning on LTP in the hippocampal slice. Methods:Mice were divided into 3 groups randomly and were exposed to hypoxia,respectively, for 4 runs (H4 group) ,1 run (H1 group) and 0 run (control group). The hippocampal slices were prepared and LTP were recoded after mice exposed to hypoxia. Schaffer lateral branch in CA3 region was simulated to record LTP of each group in CA1 region. Results :The probability of the induction of LTP in the CA1 region of H1 group was the lowest among three groups. The percentage of both A - PS and f- EPSP changes in hippocampal slices of group H0 and H4 was significantly higher than that in group H1 ( P 〈0.01 ), but no difference was seen between group H0 and H4 ( P 〉 0.05 ). Conclusion: Hypoxia inhibiting the induction of LTP and hypoxia preconditioning can recover the induction. The change of synaptic plasticity may be involved in the protection of brain of hypoxia - preconditioned mice.
