摘　　要：目的:以兔血清白蛋白和IgG的分离纯化与鉴定实验为例,阐述了血清蛋白制备与分析的过程。方法:使用了硫酸铵分级沉淀、凝胶过滤层析、DEAE离子交换层析、亲和层析、醋酸纤维薄膜电泳、SDS-PAGE、HPLC等分离纯化鉴定蛋白质的方法。结果:制备了兔血清白蛋白和γ-球蛋白(IgG),测定被分离物质的相对分子量,鉴定纯化产品的均一性。结论:该实验作为一门生物综合性大型实验,非常有利于培养学生掌握蛋白质制备与分析的综合技能。Objective：To explore the method to prepare serum protein and how to analyze the result through the experiment of extraction,purification and identification of rabbit serum albumin and IgG. Methods：Ammonium sulfate＇s fractional precipitation, gel filtration chromatography,DEAE ion exchange chromatography, affinity chromatograph, cellulose acetate membrane electrophoresis, SDS-PAGE, HPLC were used. Results：Rabbit serum albumin and IgG were prepared, the relative molecular weight was determined,and the producti homogenicity was identified. Conclusion：Through this biotic comprehensive experiment ,students will gradually master the technique of preparing protein and analyze result.
