摘 要:目的观察关节盘移位后髁突软骨中c-fos基因转录与表达水平的变化。方法32只日本大耳白兔,实验组25只用于建立关节盘前移位的动物模型,5只行模拟手术作为手术对照组,分别于术后1、2、4、8、12周处死;另2只不行手术作为空白对照。原位杂交及免疫组化法分别检测髁突软骨中c-fos基因的转录与表达水平,计算机图像分析系统计算平均灰度值。结果实验组关节盘移位1周后,髁突软骨c-fos mRNA的灰度值为57.66±4.80,C-FOS蛋白水平的灰度值为144.12±12.43,与对照组相比均有明显增高,其差异有统计学意义(P<0.05)。关节盘移位4周后,髁突软骨c-fos mRNA表达和C-FOS蛋白水平开始回落。关节盘移位8~12周后两者基本达到正常水平,髁突软骨c-fos mRNA的灰度值为16.64±0.08,C-FOS蛋白水平的灰度值为46.72±5.23,与对照组相比差异均无统计学意义(P>0.05)。结论关节盘前移位的早期髁突软骨c-fos基因转录与表达水平都明显升高,可能在关节盘移位后髁突软骨的改建过程中发挥着重要信号转导作用。Objective To evaluate gene transcription and expression of c-fos in mandibular condyle cartilage after anterior disc displacement (ADD). Methods Thirty-two healthy adult Japanese rabbits were used in this study. Twenty-five rabbits were subjected to surgical anterior disc displacement in the right temporomandibular joints, every 5 rabbits were sacrificed 1, 2, 4, 8, 12 weeks post-operation, 5 rabbits were used as sham-operative control and the other two without operation were used as normal control. The gene transcription and expression of c-fos in condyle cartilage were studied by in situ hybridization and immunohistochemical methods and measured with Image Computer Analysis Software. Results The discs displaced anteriorly were observed successfully in the experimental rabbits. 1 week after ADD, the data of staining by in situ hybridization and immunohistochemical methods were 57.66±4.80, 144.12±12.43 respectively, gene transcription and expression of c-fos increased significantly (.P〈0.05). Four weeks after ADD, the gene transcription and expression of c-fos began to decrease, 8-12 weeks after ADD, the gene transcription and expression of c-fos almost came back to the level before ADD, the data of staining by in situ hybridization and immunohistochemical methods were 16.64±0.08, 46.72±5.23 respectively, gene transcription and expression of c-fos had no significant differences (P〉0.05). Conclusion In the early stage of ADD, the level of c-fos gene expression and transcription in mandibular condyle cartilage increased significantly. It may suggest that c-fos might play an important role in the signal transduction in the remodeling of the mandibular condyle cartilage after ADD.
