间歇性低氧对少突胶质前体细胞增殖的影响

摘  要:目的:观察间歇性低氧-复氧对少突胶质前体细胞增殖的影响。方法:取分离纯化的少突胶质前体细胞,培养皿或96孔板中的细胞分别加入10mg/ml的各种细胞因子,分为常氧组和低氧组,低氧组在3%低氧环境下持续4d分别各培养1h、2h、3h、4h后取出复氧,9d后进行MTT测定或细胞计数。96孔板分组;无细胞的阴性对照,有细胞的空白对照,PDGF,bFGF,IL-6,IFN-β1,GDNF,PDGF+bFGF,PDGF+IL-6,PDGF+GDNF,bFGF+IL- 6,bFGF+GDNF,IL-6+GDNF,PDGF+bFGF+IL-6,PDGF+bFGF+GDNF,PDGF+bFGF+IL-6+GDNF共16组。培养皿分组除无细胞的阴性对照组外的15组。培养至9d。同时取出各组细胞,进行细胞计数和MTT测定。结果:除1h组外,各低氧处理组细胞数及MTT值均较对照明显增多。结论:首次报道恰当时间的间歇性低氧-复氧可促进少突前体细胞增殖。这为细胞移植治疗中枢神经系统脱髓鞘疾病及研究髓鞘形成、髓鞘再生的机理提供基础。Objective: To explore the effect of hypoxia and teoxygenation on the proliferation of oigodendrocyte precursors in vitro. Ogodendrocytes were considered as endurable to hypoxia compared with other neural cells such as neurons and astrocytes. Method: Oigodendrocyte precursors which grew on the surface of astrocytes monolayer were obtained :from new neonatal SD rat cortex which cultured in % pore plate or Petri dishes. In the control group, O2A cells were incubated in normoxic conditions (20 % O2) for 9d. In the hypoxia - reoxygenation group, O2A cells were first incubated under hypoxic conditions (3 % O2) for 1h, 2h. 3h end 4h respectively for 1 - 4d, and were then incubated in normoxic conditions till the 9th day. In the cytokine group, there were 16 subgroups as follows: negative control (no cells), control (with cells, no cytokines), PDGF, bFGF, IL - 6, IFN-β1, GDNF, PDGF + bFGF, PDGF + IL- 6, PDGF+ GDNF, bFGF + IL- 6, bFGF + GDNF, IL - 6 + GDNF, PDGF + bFGF + IL- 6, PDGF + bFGF + GDNF, PDGF + bFGF + IL - 6 + GDNF, the concentration of cytokines was 10ng/ml, On the 9th day, the effect of hypoxia and cytokines on O2A cells were testified by MTT assay and cell number calculating. Results: O2A cells increased significantly in 2h, 3h and 4h of the hypoxia- reoxygenation group. Conclusion: Hypoxia and reoxygenation can signiflcantly promote the proliferation of oligodendrocyte precursors. The mechanism should be studied in details.
