摘 要:目的探讨孕鼠叶酸缺乏(FD)对子代心脏发育过程中GSK3β基因表达的影响。方法成熟雌性SD大鼠36只随机分为实验和对照组各18只,分别喂以缺乏叶酸和添加叶酸纯合饲料。2周后与成熟SD雄性大鼠交配,分别取孕13.5、17.5d胚胎鼠及新生鼠心脏。用逆转录聚合酶链反应(RT-PCR)检测GSK3β基因mRNA表达。结果GSK3βmRNA在孕13.5、17.5d胚胎心脏及新生鼠心脏中的表达量,实验组均显著低于对照组(Pa<0.05)。结论叶酸缺乏可影响GSK3β基因表达水平,可能导致心脏发生发育中形态改变,造成心脏功能缺陷。Objective To study the expressions of glycogen synthase kinase 3β(GSK3β) genes on the heart of offspring during the development of embryo, whose mother was deficient of folic acid. Methods Thirty - six adult female SD rats were chosen and alloratad randondy into control group (18 rats) and study group (18 rats). The control group received nonfolic acid fodder and the study group received folic acid fodder for 2 weeks, Then the female rats in 2 groups copulated with normal male rats, The hearts of the 13.5 days,17, 5 days embryos and the newborns were obtained, The expressions of GSK3βgenes mRNA at the 3 periods were measured by reverse transcriptase - polymerase chain reaction(RBT - PCR). Results The expression levels of GSK315 mRNA in study group were significantly weaker than that in control group in hearts of the 13, S days, 17, 5 days embryos and the newborns( P〈0.05 ). Conclusion Changes of expression of GSK3β mRNA may be resulted from folic acid deficiency during the pregnancy, which may lead to the malformation of the heart.
