载脂蛋白E基因多态性与难治性精神分裂症关联分析

摘　　要：目的比较汉族人群难治性精神分裂症与非难治性精神分裂症患者外周血载脂蛋白(apoE)基因多态性的差异。方法采用聚合酶链反应和限制性片段长度多态性技术,检测75名正常人(以下简称对照组)、138例难治性精神分裂症患者(以下简称难治组)及97例非难治性精神分裂症患者(以下简称非难治组)的 apoE 基因型,分别比较精神分裂症患者与对照组、难治组和非难治组间的 apoE 基因多态性。结果精神分裂症患者 apoE 3/4基因型、ε_4等位基因频率(39.6%,21.9%)明显高于对照组(6.7%,3.3%),而其2/2基因型、3/3基因型和ε_3等位基因(0.0%,48.5%,72.3%)明显低于对照组(1.3%,78.7%,88.7%),P 均<0.01。难治组 apoE 3/4基因型、ε_4等位基因频率(46.4%,26.1%)高于非难治组(29.9%,16.0%),而其2/3基因型频率(4.3%)明显低于非难治组(13.4%),P 均<0.01～0.05。结论 apoE 3/4基因型和ε_4等位基因可能是精神分裂症及其难治性的危险因子之一。Objective To compare the differences between refractory and non-refractory schizophrenia in polymorphism of apolipoprotein E （apoE） in the Han nationality, Methods Seventy-five normal subjects, 138 patients with refractory schizophrenia and 97 with non-refractory schizophrenia were included in this study. The gene polymorphism of apoE in all subjects was genotyped directly with polymerase chain reaction and restriction fragment length polymorphism technique. The differences of apoE polymorphism between schizophrenia and normal subjects, refractory and non-refractory schizophrenia were compared. Results The frequencies of apoE genotype 3/4 and allele 4 in schizophrenia group （ 39. 6% , 21.9% ） were significantly higher than that in control group （6. 7% , 3.3% ） , while the frequency of apoE gene2/3, gene 3/3 and allele 3 in schizophrenia group （0. 0% ,48.5% ,72. 3% ） were lower than that in control group （ 1.3% ,78.7% ,88.7% ）. The frequencies of apoE genotype 3/4 and allele 4 in refractory schizophrenia group （46. 4% , 26. 1% ） were significantly higher than that in non-refractory schizophrenia group （29.9% ,16.0% ）, while the frequency of apoE gene 2/3 in refractory schizophrenia group （4. 3% ） were lower than that in non-refractory schizophrenia group （13.4%）. Conclusion ApoE allele 4 and genotype 3/4 may play a role in schizophrenia and refractory schizophrenia.
