摘  要:目的观察海马移植脐血干细胞对血管性痴呆(VD)大鼠脑内Ach含量及AchE活性的影响。方法分离正常分娩胎儿的脐带血单个核细胞,以贴壁培养法得到间质干细胞(MSCs),采用RT-PCR方法检测Nestin及musashi-1。制作VD大鼠模型,8~10d后行脐血干细胞海马移植。移植4周后,检测大鼠的学习记忆能力,并检测其脑内乙酰胆碱(Ach)含量及乙酰胆碱脂酶(AchE)活性。结果脐血干细胞在分离后48h高表达Nestin及musashi-1;海马移植脐血干细胞4周后,治疗组与模型组比较,Ach含量及AchE活性明显升高(P<0.01),大鼠学习记忆能力增强。结论脐血中表达神经干细胞(NSCs)的标志物,海马移植脐血干细胞能明显改善VD大鼠胆碱能系统的功能,调节脑内Ach生理代谢,增强学习记忆能力,从而达到治疗VD的目的。Objective To observe the effect of transplanted human cord blood stem cells in VD rats, by detecting the activity of acetylcholinesterase(AchE) and content of Ach. Methods Human cord blood stem cells were prepared from normal placenta after fullterm normal delivery and isolated by standard Ficoll-Hypaque technique. Reverse transcriptase pol- ymerase chain reaction(RT-PCR) for nestin and musashi-1 antigen were performed to confirm nestin and musashi-1 antigen expression in the human cord blood cells. Modified 2-vessel occlusion(2-VO) was adopted to make a rat model of VD. 8-1Od after operation ,the treatment group were administered with human cord blood cells 3 x 106/ixL through hippocampus transplantation. After 4 weeks, the activity of acetylcholinesterase(AchE) , content of Ach in hippocampi and the AAR ratio of rat model were detected. Results Nestin and musashi-1 antigen both showed the highest expression in human cord blood 48 h after isolation, and compared with the control group,the hippocampus Ach content and AchE activity increased significantly in the treatment group( P 〈 0. 01 ). Conclusions NSCs markers expressed in human cord blood; function of cholinergic system can be improved significantly by transplanted human cord blood stem cells, which may be the mechanism of VD therapy.
