摘  要:目的探讨角膜碱烧伤后,碱性成纤维细胞生长因子(b-FGF),血管内皮生长因子(VEGF)在大鼠角膜中的表达与角膜新生血管化的关系。方法采用1mol/L的氢氧化钠溶液烧伤35只Sprague-Dawleg(S-D)大鼠角膜,建立角膜碱烧伤动物模型;碱烧伤后不同时间的形态学分析来评价角膜新生血管的情况,用免疫组织化学染色方法和计算机图像分析系统检测大鼠角膜烧伤后不同时间点b-FGF及VEGF在角膜中的表达。结果大鼠角膜烧伤后2db-FGF及VEGF开始增高,伤后7d达到高峰,然后2种因子的表达下降,14d后显著下降,碱烧伤后角膜新生血管与两种因子的表达呈明显的平行关系。结论碱烧伤后,大鼠角膜b-FGF,VEGF的表达与新生血管的形成有相关性,并在其中发挥重要的作用。Objective To study the relationship between corneal neovascularization and expression of basic fibroblast growth factor(b-FGF) and vescular endothelial growth factor (VEGF) in mouse cornea with alkali burn. Methods Corneal neovascularization was induced by NaOH cauterizing the surface of cornea in 35 S-D rats. The corneas were enucleated for histopathological examination;the areas of neovascularization of different time points were calculated. The expression of b-FGF and VEGF were studied by immunohistochemistry and analyzed by computer medical image analysis system. Results The expression of b-FGF and VEGF elevated at the 2nd day,peaked at the 7th day,decreased after the 7th day, dropped to lowest level at the 14th day. It' s paralleled with the neovascularization. Conclusions The expression of b-FGF and VEGF may play an important role in CNV in mouse cornea with alkali burn.
