摘 要:目的:探索水苏糖酸水解制备甘露三糖的方法。方法:通过HPLC定量检测,采用正交试验设计,考察温度、pH值、时间三个影响因素,来确定水苏糖的酸水解工艺;并经柱层析分离得到甘露三糖。结果:在本实验条件下,温度和时间对水苏糖酸水解影响不显著(P<0.05),而酸度对其有显著性影响(P>0.05),确定最佳反应条件为90℃、24 h、pH 2.5。结论:水苏糖酸水解制备甘露三糖的方法可行。Objective: To optimize the processing of mannatriose preparation from stachyose by acidolysis. Methods: An orthogonal experiment was performed to select a better condition of acidolysis. Fructoes and stachyose was determined by HPLC. Results: Among the factors set in the present work, temperature and time have little influence on the acidolysis, while the effect of pH value was significant (P 〈0. 05). The best processing was selected as heating stachosy solution for 12h at pH 2. 5 and 90℃. Conclusion: It is practicable to prepare manninotriose from stachyoses by acidolysis.
