吗啡依赖及戒断大鼠相关脑区Bcl-2的表达研究

摘　　要：目的:明确吗啡依赖及戒断两种状态时大鼠相关脑区Bcl-2的表达特征。方法:将18只Sprague-Dawley雄性大鼠随机分为吗啡依赖组、吗啡戒断组和空白对照组,每组6只。采用递增法皮下注射吗啡建立大鼠吗啡依赖模型,纳络酮注射戒断,应用免疫组化和地高辛标记寡核苷酸探针原位杂交技术检测三组大鼠不同脑区Bcl-2的表达水平。结果:连续给予吗啡5天及纳络酮戒断后,中脑腹侧被盖区、海马、蓝斑、前额叶皮质脑区Bcl-2及其mRNA较对照组显著降低(P<0.05),而吗啡戒断组伏隔核脑区内Bcl-2及其mRNA的表达较依赖组显著增加(P<0.05),结论:吗啡依赖及戒断大鼠Bcl-2蛋白及其mRNA的表达在不同脑区有选择性的改变,可能是吗啡依赖及戒断的分子机制之一。Objective：To investigate the expression of Bcl-2 in rat brain during dependence and withdral state. Methods：The rats were rendered dependent by subcutaneous injections of increasing doses of morphine from 10 to 80 mg/kg for 5 consecutive days. The rats as controls were administered with same volume of normal saline. The expression of Bcl-2 mRNA was assayed with digacin-labelled Bcl-2 cDNA probe by in situ hybridization and the levels of Bcl-2 protein were measured by immunohistochemistry. Results：The evels of Bcl-2 and their mRNA in prefrontal cortex,locus coeruleus, hippocampus and ventral tegmental area significandy decreased as eomparied with that in control rats after chronic administration of morphine and naloxone administration.However, Bcl-2 and their mRNA in nucleus accumbens of rats in withdral state were markedly higher than that in nucleus aecumbens of morphine dependent rats. Conclusion：Chronic exposure to morphine appears to induce modification of the expression of Bcl-2 mRNA and their protein of rats in the rat brain,which contributes to the development of morphine dependence .
