摘  要:目的:探讨高浓度葡萄糖作用于脐静脉内皮细胞后,对脐静脉血管内皮细胞系的基质金属蛋白酶(MMP-2)表达、超微结构及细胞周期的影响,为阐明高血糖在糖尿病血管病变、糖尿病视网膜病变的早期发病中作用与机制提供依据。方法:在高浓度葡萄糖条件下培养人脐静脉内皮细胞,采用RT-PCR、Western-blot法检测细胞基质金属蛋白酶(MMP-2)mRNA和蛋白表达情况、电镜观察超微结构及流式细胞仪检测细胞周期的变化。结果:高浓度葡萄糖可使内皮细胞MMP-2表达增加、细胞增生活跃,细胞出现肿胀,体积增大,细胞质溶解、核溶解,异染色质边集,内质网扩张,出现固缩线粒体、环形线粒体,髓样结构等改变。结论:高糖诱导的MMP-2mRNA和蛋白表达增加,可能在糖尿病视网膜病变的早期发病中起重要作用。Objective:To examine the effects of glucose on the expression of matrix metalloproteinases-2 (MMP-2)and the changes of ultramicrostructure,cell cycle in cultured human umbilical vein endothelial cells. Methods:Human umbilical vein endothelial cells( hUVEC )were incubated in a culture medium with l 1.2mmol/L,16.8mmol/L and 33.6mmol/L glucose concentrations for 24h,48h,72h respectively. The expression of MMP-2 was measured by RT-PCR and Western-blot. Results:When D-glucose concentrations rose,hUVEC exposed to high glucose concentration(11.2mmol/L,16.Smmol/L and 33.6mmol/L)increased in cell expression of MMP-2 after 48 h and 72 h of exposure,as compared with those cultured in medium with a low glucose concentration (5.6mmol/L). And cell cycle changed too. Conclusion:High concentration of glucose can arrest the proliferative response and show that even a short-term exposure of endothelial cells( ECs )to high glucose concentration leads to their activation associated with increased expression of MMP-2 and that this effect may appear more significantly along with time of exposure to high concentration of glucose.
