表皮生长因子对体外建株瘤细胞的促增殖作用及其对瘤细胞顺铂敏感性的影响

摘　　要：本文采用３，４，５－二甲基噻唑－２，５－二苯基－四唑溴化物（ＭＴＴ）比色法及乳酸脱氢酶释放法，研究了表皮生长因子（ＥＧＦ）对人骨髓红白血病细胞系（Ｋ５６２）、人Ｂ淋巴细胞白血病细胞系（Ｒａｊｉ）、人宫颈癌细胞系（Ｈｅｌａ）的促增殖作用及其对３种瘤细胞顺铂（ＣＤＤＰ）敏感性的影响。结果显示，ＥＧＦ能够促进Ｒａｊｉ、Ｈｅｌａ细胞增殖，对Ｋ５６２细胞的生长没有促进作用。ＥＧＦ与ＣＤＤＰ同时接触瘤细胞，不能增高瘤细胞对ＣＤＤＰ的敏感性，但提前２４小时接触瘤细胞，Ｒａｊｉ、Ｈｅｌａ细胞对ＣＤＤＰ敏感性明显增高；Ｋ５６２细胞的敏感性则未见增高。表明ＥＧＦ增高肿瘤细胞ＣＤＤＰ敏感性可能与其提前激活瘤细胞表面的ＥＧＦ功能性受体有关。揭示ＥＧＦ激活的信号传导通路在ＣＤＤＰ细胞毒作用中起调节作用。$$$$ Hong Xin,et al Department of Hygiene,Dalian Medical UniversityAbstract 3,4,5 dimethyflthiazol 2,5 disphenyl tetraolium bromide(MTT)assay and lactic dehydrogenase release into the culture medium were used to study effect of epidermal growth factor(EGF)on proliferation of human erythroleukemia cell line(K 562 ),human B lymphocytic cell line(Raji),human cervix carcinoma cell line(Hela)and on sensitivity of these cells to cisplatin (CDDP).Our results showed that EGFpromoted proliferation of Raji,Hela cells,but it had no effect on K 562 cells.Cellular sensitivity to CDDP was not enhanced by simultaneous administration of EGF ,but sensitivity of Raji,Hela cells to CDDP was enhanced by previous administration of EGF,sensitivity of K 562 cells to CDDP was not enhanced.Our findings indicate that promoting proliferation of EGF could be the base of enhanced sensitivity to CDDP;EGF could not enhance sensitivity to CDDP if only cells are previously treated with it,EGF activated signal transduction pathway could modulate toxicity of cells to CDDP.
