摘 要:目的研究钾通道阻断剂四乙铵(TEA)对卵巢癌细胞(SKOV3)凋亡的影响。方法将浓度为2×104/ml的卵巢癌细胞分别设对照组和不同浓度(1、5、10、20mmol/L)TEA组。将TEA作用于卵巢癌细胞,用MTT法检测细胞活性,采用Hoechest33258染色检测细胞凋亡,同时通过碘化丙啶(PI)染色,采用流式细胞仪检测细胞凋亡比率。结果在TEA浓度为1、5、10、20mmol/L时,对SKOV3细胞增殖的抑制率分别为8·61%、18·86%、46·63%和65·22%,表明TEA对SKOV3细胞的增殖有抑制作用并呈现明显的剂量依赖性,当TEA浓度=5mmol/L时,对SKOV3细胞增殖的抑制效应明显;Hoechst33258染色结果显示,TEA能诱导SKOV3细胞凋亡;流式细胞仪检测结果表明,SKOV3细胞经TEA浓度为1、5、10、20mmol/L处理后48h,其细胞凋亡率分别为9·23%、21·57%、54·22%和71·06%,与对照组(2·08%)比较,差异有显著性(P<0·01),提示TEA可诱导SKOV3细胞凋亡,且TEA促进SKOV3细胞凋亡作用具有一定的剂量依赖性。结论钾通道对SKOV3细胞增殖具有重要调控作用,钾通道阻断剂TEA阻断钾通道后可促进细胞凋亡。Objective To study the effects of potassium channel blocker, tetraethylammonium (TEA) on the proliferation and apoptosis of ovarian cancer cell line SKOV3. Methods SKOV3 cells were treated with TEA. The effect of TEA on proliferation of SKOV3 cells was assessed by MTT assay. Hoechst 33258 fluorescein staining and flow cytometry (FCM) were employed in detection of SKOV3 cells apoptosis. Results TEA (1, 5, 10 and 20mmol/L) can significantly inhibit the proliferation of SKOV3 cells, the inhibitory rates were 8. 61%, 18. 86%, 46. 63% and 65. 22%, respectively (P〈0. 05, compared with control group). The results showed that TEA dose- dependently inhibited the proliferation of SKOV3 cell. By using Hoechst 33258 method, it was found that TEA (in 1, 5, 10 and 20mmol/ L) may induce and obviously increase the number of apoptosis of SKOV3 cells. Examination with flow cytometer (FCM) also revealed that, in the concentrations of 1, 5, 10 and 20 mmol/L, TEA can markedly promote the apoptosis of SKOV3 cells, the apoptotic rates were 9. 23%, 21. 57%, 54. 22% and 71. 06%, respectively (P〈0. 01, compared with control group). It was suggested that TEA could inhibit the proliferation of SKOV3 cells and promote the apoptosis of SKOV3 cells in dose-dependent manners. Conclusions Potassium channel play an important role in regulating proliferation of SKOV3 cells, potassium channel blocker, TEA could induce SKOV3 cell apoptosis.
