摘 要:目的:观察人甲状旁腺激素1-34(human parathyroid hormone1-34,hPTH1-34)对牙本质的形成和矿化的影响,探讨PTH影响牙本质形成和矿化的机制。方法:切除大鼠甲状旁腺,建立牙本质发育不全的动物模型后,再人工给予不同剂量的hPTH1-34,检测血钙水平的变化,42d后处死大鼠,取下颌切牙制作切片,HE染色,光镜观察并进行图像分析。结果:切除甲状旁腺的大鼠血钙水平下降,牙本质矿化缺陷。在给予hPTH1-34皮下注射后可以检测到血钙水平上调,光镜观察和图像分析结果发现牙本质的形成和矿化发生逆转,10nmol/kg组和20nmol/kg组作用效果优于5nmol/kg组(P<0.05)。结论:PTH可能通过调节血钙或其自身直接作用影响牙本质的形成和矿化。Objective:To study the effects of parathyroid hormone(PTH) upon dentinogenesis by investigating the effects of hPTHI-34 on the development and calcification of parathyroidectomy rats' dentin. Methods: hPTH1-34 were injected into SD rats at different dosages after parathyroidectomy. Six weeks later lower incisors were dissected and prepared for light microscopy and image analysis. Results:The concentration of serum calcium decreased after parathyroidectomy, while increased after being injected hPTH1-34. The results of light microscopy and image analysis revealed the enlargement of the predentinal tubules and the presence of mineralization defects in the PTX group (P 〈0.05). These defects were improved after injection of hPTH1-34. The groups of 10 nmol/kg and 20 nmol/kg affected the dentin more than group of 5 nmol/kg( P 〈 0.05 ). Conclusion: The development and mineralization of dentin can be affected by PTH.
