食管鳞癌中多药耐药相关蛋白表达与其病理特征的关系

摘　　要：目的探讨多药耐药相关蛋白(multidrug resistance-associated protein,MRP)在食管鳞癌组织的中表达。方法运用免疫组化方法检测MRP蛋白在81例食管鳞癌组织中的表达;对其中的32例运用RT-PCR技术检测MRP mRNA的表达。结果食管癌组织MRP蛋白的表达增高(54%),明显高于正常组织中MRP的表达(35%)(P<0.05)。MRP蛋白在深层浸润组的表达(61%)高于浅层浸润组(35%)(P<0.05);MRP蛋白在有淋巴结转移组的表达(70%)明显高于无淋巴结转移组(44%)(P<0.05);MRP蛋白的表达与癌组织的分化程度无关(P>0.05)。MRP mRNA在食管正常黏膜及食管鳞癌组织中的表达差异无统计学意义(P>0.05),且与癌组织的分化程度、浸润深度、淋巴结转移等无关(P>0.05)。结论MRP的高表达可能与食管鳞癌的发生发展有关,MRP蛋白可作为判断食管鳞癌恶性程度及预后的一个参考指标。Objective To investigate the expressions of muhidrug resistance-associated protein（MRP） in esophageal carcinoma. Methods The expressions of MRP protein were detected by immunohistochemical staining in 81cases with esophageal carcinoma tissues. In 32 of the cases, MRP mRNA expressions were detected by RT-PCR. Results The expressions of MRP protein were higher significantly in carcinoma tissues than those in normal tissues （P 〈 0.05 ）. The expressions of MRP in the deeper invasion group and the group with lymph node metastasis were significantly higher than those in the superficial invasion group and the group without lymph node metastasis（P 〈 0.05 ）. It was not observed that MRP protein expressions were related to carcinoma differentiation （P 〉 0.05 ）. The expressions of MRP mRNA showed no difference between esophageal normal tissues and carcinoma tissues（P 〉 0.05 ）. It was not found that MRP mRNA expressions were related to carcinoma differentiation, the depth of invasion and lymph node metastasis（P 〉 0.05 ）. Conclusions It indicated that the overexpressions of MRP protein might involve in esophagus carcinogenesis, progression and the formation of metastasis. MRP protein could be an effective factor in judging the malignant degree in esophageal carcinoma.
