摘 要:目的观察乳腺癌组织、癌旁相对正常乳腺组织中抑癌基因p53和凋亡调节蛋白Bcl-2的表达,探讨Bcl-2和p53在乳腺癌发生、发展过程中的作用及其相互关系。方法收集21例手术切除的人乳腺癌组织和癌旁相对正常乳腺组织,用免疫组织化学方法对不同乳腺组织标本进行检测。结果Bcl-2在相对正常乳腺组织、非浸润性乳腺癌组织、浸润性乳腺癌组织中有不同程度的表达,其强阳性表达率分别为19.0%、41.7%、55.5%;p53在相对正常乳腺组织、非浸润性乳腺癌组织、浸润性乳腺癌组织中有不同程度的表达,其强阳性表达率分别为0%、50.0%、66.6%;而Bcl-2与p53表达之间存在显著相关性。结论Bcl-2蛋白在乳腺癌组织中高表达与肿瘤的浸润有关;p53在乳腺癌组织中的过度表达,可能抑制乳腺癌细胞的凋亡发生,促进肿瘤的生长,与乳腺癌进展有关。p53及Bcl-2基因在乳腺癌发生发展过程中发挥着既独立又协同的作用。Objective To investigate the expressions and mutual relationship of tumor gene p53 and apoptosis regulating gene Bcl-2 in breast cancer and to discuss the relationship between apoptosis gene and developing of breast cancer. Methods The human breast cancer tissues and relatively normal breast tissues were collected from surgical operation and the samples of 21 patients with breast cancer were analyzed by immunocytochemistry. Results The positive rate of Bcl-2 was 19.0% normal breast tissues,41.7% in intraductal carcinomas,55.5% in invasive carcinomas. Higher Bcl-2 expressions were observed in breast cancer than those in normal breast tissues. Furthermore, Bcl-2 posi- tivity in invasive carcinomas was significantly higher than that in no invasive carcinomas . No p53 nuclear accumulation was observed in normal breast tissues. The positive rate of p53 was 50.0% in no invasive carcinomas, 66.6% in invasive carcinomas. Higher p53 expressions were observed in breast cancer tissues higher than those in normal breast tissues. Furthermore, p53 positivity in invasive carcinomas was significantly higher than that in intraductal carcinomas. Conclusions High levels of Bcl-2 expressions in breast cancer have related to tumor aggressiveness, p53 expressions may occur early in breast cancer development and increases during progression. The high expressions of p53 gene in breast cancer were closely associated with the tumor development, p53 and Bcl-2 may play both independent and synergetic roles in the course and development of breast cancer.
