摘  要:尽管树突状细胞(dendritic cell,DC)是机体内最有效的抗原提呈细胞(antigen presenting cell,APC),但由于逃逸机制的存在,基于DC的肿瘤疫苗效果并不如人意。因此,采用一定方法增强DC提呈抗原的能力是提高疫苗效能的关键。本研究中,采用RNA干扰技术(RNA interference,RNAi),沉默DC中细胞因子信号转导抑制因子1(suppressor of cyto- kine signaling 1,SOCS1)的表达,使之提呈抗原的能力显著提高,从而有效激发针对肿瘤的特异免疫应答,疫苗的治疗效果得以明显增强。Although dendritic cells(DC) are the most potent professional antigen-presenting cells to activate the innate and adaptive immunity against lung cancer, however, on account of the presence of immune escape mechanism, the therapeutic effect of vaccine based on DC is only of limited success in the clinic trails. The major challenge is to find a means to overcome the inhibitory mechanism and to provoke an effective T cell response to antigens expressed by tumor cells, in which improvement of the ability in antigen presentation of DC vaccine may be the main point to enhance its therapeutic effect. In the present study, we had adopted the RNA interference(RNAi)technique to inhibit the expression of suppressor of cytokine signaling-1 (SOCS1)in order to silence the effect of SOCS1 of suppressing the activities of antigen presenting ability of DC and thus efficiently activating the specific anti-tumor immunity. In this way, the therapeutic effect of vaccine based on DC are effectively enhanced.
